-abstract: "Narrative to introduce the Nextstrain-TrepoGen project (https://github.com/Integrative-Transcriptomics/Nextstrain-TrepoGen) during ISMB/ECCB2025: With 7 million cases of infection in 2020, syphilis and other treponematoses are a global public health concern. The WHO states that progress in fighting this sexually transmitted diseases has plateaued. Although penicillin treatment is inexpensive and effective, vaccination is considered key to eliminate the disease. _Treponema pallidum_ (Tp), the causative agent of syphilis, is considered a “stealth pathogen” due to its poorly antigenic and low-inflammatory surface. Progress has been made in identifying the structure and function of Tp outer membrane proteins (OMPs), some of which are promising vaccination targets. With this project we want to democratize access to genomic diversity data of relevant Tp subspecies by developing and maintaining unified Nextstrain5 datasets for epidemiological surveillance, comparative evolutionary analysis, and facilitated data sharing. We are placing a special emphasis on tracking OMP diversity relevant for vaccine design."
